I was lucky and took the upgrade exams since I was already a MCSE for NT, 2000 and 2003. I went straight to the MCITP: Enterprise Administrator cert with the upgrade and Pro exams along with the Vista cert I already had.
The best thing you can do is read these books cover to cover. Then setup a lab in a VMWare (or whatever your virtualization preference is) environment. I would do at least 2 or 3 domain controllers in seperate subnets to emulate multiple sites. Knowing how Sites and Services work inside and out will be key to passing these exams. Pay specific attention to replication topics, KDC, Site Links and Site Link Bridges. Also, be sure you know the FSMO roles, what they do and where to properly place those roles. Understanding DNS is of course a huge part of these exams as everything in Windows requires DNS, especially Active Directory.

From there, the MCITP: Enterprise Messaging exams are focused around Exchange 2010, you will need 3 exams to get that cert. They are not easy by any means and require a lot of exposure to large scale Exchange environments to be properly prepared for those exams. Know your Powershell cmdlets!
I’m working towards getting a slot in the Microsoft Certified Master: Exchange 2010 program sometime next year.
